Heart Failure Critical Role for Stromal Interaction Molecule 1 in Cardiac Hypertrophy

چکیده
Background—Cardiomyocytes use Ca not only in excitation-contraction coupling but also as a signaling molecule promoting, for example, cardiac hypertrophy. It is largely unclear how Ca triggers signaling in cardiomyocytes in the presence of the rapid and large Ca fluctuations that occur during excitation-contraction coupling. A potential route is store-operated Ca entry, a drug-inducible mechanism for Ca signaling that requires stromal interaction molecule 1 (STIM1). Store-operated Ca entry can also be induced in cardiomyocytes, which prompted us to study STIM1-dependent Ca entry with respect

Cardiac Stim1 Silencing Impairs Adaptive Hypertrophy and Promotes Heart Failure Through Inactivation of mTORC2/Akt Signaling.
BACKGROUND Stromal interaction molecule 1 (STIM1) is a dynamic calcium signal transducer implicated in hypertrophic growth of cardiomyocytes. STIM1 is thought to act as an initiator of cardiac hypertrophic response at the level of the sarcolemma, but the pathways underpinning this effect have not been examined. METHODS AND RESULTS To determine the mechanistic role of STIM1 in cardiac hypertro...

متن کاملEssential Role of Smooth Muscle STIM1 in Hypertension and Cardiovascular Dysfunction.
OBJECTIVES Chronic hypertension is the most critical risk factor for cardiovascular disease, heart failure, and stroke. APPROACH AND RESULTS Here we show that wild-type mice infused with angiotensin II develop hypertension, cardiac hypertrophy, perivascular fibrosis, and endothelial dysfunction with enhanced stromal interaction molecule 1 (STIM1) expression in heart and vessels. All these pat...
متن کاملSTIM1/Orai1-mediated SOCE: current perspectives and potential roles in cardiac function and pathology.
Store-operated Ca²⁺ entry (SOCE) is critical for Ca²⁺ signaling in nonexcitable cells; however, its role in the regulation of cardiomyocyte Ca²⁺ homeostasis has only recently been investigated. The increased understanding of the role of stromal interaction molecule 1 (STIM1) in regulating SOCE combined with recent studies demonstrating the presence of STIM1 in cardiomyocytes provides support th...
متن کاملLactobacillus Paracasei Has Anti-Inflammatory Effect on the Heart Failure Induced by Isoproterenol in Rats
Background: Heart failure (HF) has become one of the most prevalent cardiovascular problems worldwide. Considering the beneficial effects of probiotics on human health, we aimed to investigate the anti-inflammatory effect of oral administration of Lactobacillus paracasei in HF induced by isoproterenol. Methods: Forty Wistar male rats weighing 80g on average were randomly assigned to five grou...
